Gentamicin is an aminoglycoside antibiotic, which has a bactericidal mode of action binding to the ribosome and various ribozymes to inhibit protein synthesis. Gentamicin is a hydrophilic molecule and is considered to have limited capacity to penetrate eukaryotic cells. Administered intravenously or intramuscularly, gentamicin is used to treat many bacterial infections, particularly gram-negative organisms such as meningitis, and infections of the blood, abdomen, lungs, skin, bones, joints and urinary tract, and is often used to treat severe cases of tularemia and the plague. The effect of gentamicin is mediated through the production of reactive oxygen species (ROS), which causes destablization of iron-sulfur (Fe-S) clusters and generates toxic, bactericidal compounds through the Fenton reaction. Gentamicin may cause kidney, nerve and severe hearing problems. More common side effects include nausea, headache, fever, rash and hoarseness. [PMID:26503658] [MedlinePlus Drug:a682275]
