Porcine Epidemic Diarrhea Virus (PEDv) is a large-enveloped RNA virus, which is a member of the genus Alphacoronavirus within the Coronaviridae family. The complete PEDv genome consists of 5 gene segments: open reading frame 1 (ORF1), spike (S), envelope (E), membrane (M), and nucleocapsid (N). It is encapsulated by a single N protein. The S protein contributes to the stimulation of neutralizing antibodies in the infected host. Its association to adapted growth in vitro and attenuation in vivo lends itself to use for genetic comparison among PEDV isolates and effective vaccines. The M protein is required for assembly and can also induce the production of virus-neutralizing antibodies. The E protein contributes to coronavirus budding. The N protein has functions associated with viral replication and pathogenesis.
PEDv causes viral enteritis, which leads to the destruction of villus enterocytes in pigs of all ages. It is spread by the ingestion of feces from infected pigs, resulting in vomiting, watery diarrhea, dehydration, and high mortality, especially in nursery piglets. Disease caused by PEDv is clinically similar to that caused by Transmissible gastroenteritis coronavirus (TGEV) PMID: 26689811
At least two PEDv strains have been identified in the USA: the US PEDv prototype and S INDEL variant. The INDEL variant PEDv strain has insertions and deletions (INDELs) in the spike (S) gene. PMID: 25279722 The INDEL strain is related to milder clinical signs than prototype strains. PMID: 24750580 PMID: 26748596 The genomic differences in the S gene between the US PEDv prototype and INDEL variant may be responsible for the virulence differences. Potential targets when testing for PEDv RNA include the nucleocapsid protein (N) gene and the ~2500 bp fragment of the S-gene. Source: Regenstrief LOINC
