Estriol (also oestriol) is one of the three main estrogens produced in humans. It is only produced in large amounts during pregnancy as it is made by the placenta. Levels of estriol in non-pregnant women do not change much after menopause, and levels are not significantly different from those in men. Estriol levels can be a good indicator of the health of the fetus. DHEA is produced by the adrenal cortex of the fetus, this is converted to estriol by the placenta. Low levels during pregnancy may indicate a problem with the development in the child.

Saliva is produced primarily by the parotid, submaxillary, and sublingual glands. It is secreted at the rate of 0-3 milliliters per minute which is affected by various factors including anxiety, hydration and hunger. Oral fluid is broadly used to test for drugs of abuse in employment and forensic settings, and is especially useful since it is difficult to adulterate, is quick and is non-invasive. Oral fluid is used to test a variety of analytes, including to test alcohol, HIV and other antibodies, therapeutic drugs and steroids, but its primary use is to test for drugs of abuse including amphetamines, cocaine and metabolites, opioids, methadone, cannabis and heroin.
