Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. Erythrocytes or red blood cells (RBCs) are the cells in the circulation that carry oxygen to and remove carbon dioxide from the tissues throughout the body. They are produced in the bone marrow in response to erythropoietin where they transition through six stages over a seven day period. When they are released into the circulation, their nucleus has been extruded and they measure 6-8 microns in diameter. The lifespan of RBCs is about 120 days. When the RBC ages the cell membrane becomes less pliable and tears as they cell travels through the micro vessels of the body. The damaged RBCs are removed from the circulation by the spleen. Variations in number, shape and size of RBCs are indicative of many diseases and disorders. There are many factors that may lead to decreased numbers of RBCs including hemorrhage, hemolysis, iron or vitamin deficiency, marrow failure and more. Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. A lymphocyte is a type of white blood cell in the vertebrate immune system.
A lymphocyte count is usually part of a peripheral complete blood cell count and is expressed as percentage of lymphocytes to total white blood cells counted. A general increase in the number of lymphocytes is known as lymphocytosis whereas a decrease is lymphocytopenia.
An increase in lymphocyte concentration is usually a sign of a viral infection (in some rare case, leukemias are found through an abnormally raised lymphocyte count in an otherwise normal person).
A low normal to low absolute lymphocyte concentration is associated with increased rates of infection after surgery or trauma.

Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. The change of cells from benign to malignant behavior is called malignant transformation. Usually this diagnosis is made by pathologic examination of tissues. However, often the physician during the history and physical examination can find that there have been changes in size or patient sensation and upon direct examination that there has been a change in the lesion itself. Malignant transformation is a synonym for progression to malignancy. Transformation may take place as a primary or a secondary process. Although malignant transformation may occur because of changes within the cell, it can be induced by inorganic toxic substances such as cadmium or arsenite and organics such as tobacco-specific nitrosamines.

A megaloblast is an unusually large erythroblast that can be associated with pernicious anemia (Vitamin B12 deficiency) and folic acid deficiency (collectively called megaloblastic anemia). It is differentiated from a normoblast, which is a normally-sized erythroblast. This kind of anemia leads to macrocytes (abnormally large red cells). The cause of this cellular gigantism is an impairment in DNA synthesis which delays nuclear maturation and cell division. Because RNA and cytoplasmic elements are synthesized at a constant rate despite the cells impaired DNA synthesis the cells show nuclear-cytoplasmic asynchrony.

The identification of mature and immature granulocytes, including myelocytes and metamyelocytes, in blood and bone marrow is fundamental to the diagnosis of haematological disorders. A myelocyte is a young cell of the granulocytic series, occurring normally in bone marrow, but not in circulating blood (except when caused by certain diseases). Myelocytes are derived from promyelocytes and give rise to metamyelocytes. When stained with the usual dyes, the cytoplasm in myelocytes is distinctly basophilic and relatively more abundant than in myeloblasts or promyelocytes. Numerous cytoplasmic granules are present in the more mature forms of myelocytes. Neutrophilic and eosinophilic granules are peroxidase-positive, while basophilic granules are not. The nucleus of a myelocyte is fairly regular in contour (not indented), and seems to be 'buried' beneath the numerous cytoplasmic granules. If the nucleus were indented, it is likely be a metamyelocyte.

A monocyte is a mononuclear leukocyte, intimately involved in the initiaton of an immune response. It also protects against blood-borne pathogens and migrates quickly to sites of infection in the tissues. Monocytes are usually identified in stained smears by their large bilobed nucleus. They are produced by the bone marrow from hematopoietic stem cell precursors called monoblasts. The monoblast is the first stage of monocyte maturation. The developmental stages (4 subtypes) of the monocytes are: monoblast, promonocyte, immature monocyte, and mature monocyte.

The myeloblast is a unipotent stem cell, which will differentiate into one of the effectors of the Granulocyte series.
These cells descend from the primitive reticulum cells, which are found in the stroma of the marrow. There is also an intermediate phase between the myeloblast and these primitive reticulum cell, namely the hemocytoblast. At this time several developing blood cell lines are available, like erythropoiesis and thrombopoiesis. The granulopoiesis is regulated by humoral agents, like Colony Stimulating Factor and Interleukin 3.
Most common problem with malfunctioning myeloblasts is the acute myeloblastic leukemia. The main clinical features are caused by failure of the hemopoiesis with anemia, hemorrhage and infection as result. There is a progressive accumulation of leukemic cells, because some blast progenitor cells renew themselves and will have a limited differentiated division. Key mediators are the cell-density and Colony Stimulating Factor(CSF). A few acute myeloblastic leukemia can be initiated by earlier hematologic disorder, like pancytopenia, hypoplasia of the bone marrow.

The identification of mature and immature granulocytes, including myelocytes and metamyelocytes, in blood and bone marrow is fundamental to the diagnosis of haematological disorders. A myelocyte is a young cell of the granulocytic series, occurring normally in bone marrow, but not in circulating blood (except when caused by certain diseases). Myelocytes are derived from promyelocytes and give rise to metamyelocytes. When stained with the usual dyes, the cytoplasm in myelocytes is distinctly basophilic and relatively more abundant than in myeloblasts or promyelocytes. Numerous cytoplasmic granules are present in the more mature forms of myelocytes. Neutrophilic and eosinophilic granules are peroxidase-positive, while basophilic granules are not. The nucleus of a myelocyte is fairly regular in contour (not indented), and seems to be 'buried' beneath the numerous cytoplasmic granules. If the nucleus were indented, it is likely be a metamyelocyte.

Hematopoietic stem cells (HSC) are stem cells found in the bone marrow. HSC are the precursor cells which give rise to all the blood cell types of both the myeloid and lymphoid lineages. This includes monocytes and macrophages, neutrophils, basophils, eosinophils, T-cells, B-cells, NK-cells, microglia, erythrocytes (red blood cells), megakaryocytes (e.g. platelets), and dendritic cells. As stem cells, they are defined by their ability to form multiple cell types (multipotency) and their ability to self-renew. Multipotency: Individual HSC have the ability to give rise to any of the end-stage blood cell types. During differentiation, daughter cells derived from HSC undertake a series of commitment decisions to one or the other lineages. Intermediate cells become progressively more restricted in their lineage potential, until eventually lineage-committed end stage cells are generated.

Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. A normoblast is an immature, nucleated erythrocyte that still retains a cell nucleus. It is the immediate precursor of a normal erythrocyte. The term normoblast is sometimes used as a synonym for erythroblast, but at other times it is considered a subcategory. In the latter context, there are two types of erythroblasts: 1) "normoblasts" develop as expected and 2) "megaloblasts" are unusually large erythroblasts that can be associated with pernicious anemia and folic acid deficiency (collectively called megaloblastic anemia).

An osteoblast (from the Greek words for "bone" and "germ" or embryonic) is a mononucleate cell that is responsible for bone formation. Osteoblasts produce osteoid, which is composed mainly of Type I collagen. Osteoblasts are also responsible for mineralization of the osteoid matrix. Bone is a dynamic tissue that is constantly being reshaped by osteoblasts, which build bone, and osteoclasts, which resorb bone. Osteoblast cells tend to decrease as individuals become elderly, thus decreasing the natural renovation of the bone tissue.

Plasma cells (or plasmocytes) are cells of the immune system that secrete large amounts of antibodies. They differentiate from B cells upon stimulation by antigen-activated CD4+ lymphocytes (or T lymphocytes) . They are found in the medulla of the lymph node, in the bone marrow and in the red pulp of the spleen. The differentiation of B cells into plasma cells is accompanied by many morphological changes that allow the cell to synthesize and secrete large amounts of antibodies.

A proerythroblast (or rubriblast, or pronormoblast) is the earliest of four stages in development of the normoblast.
In histology, it is very difficult to distinguish it from the other "-blast" cells (lymphoblast, myeloblast, monoblast, and megakaryoblast). The cytoplasm is blue in an H&E stain, indicating that it is basophilic.
Proerythroblasts arise from the CFU-e (colony-forming unit erythroid) cells, and give rise to basophilic erythroblasts. In the mouse, proerythroblasts are large committed progenitors that express high levels of transferrin receptor (iron acquisition receptor), the erythropoietin receptor (EpoR), some c-Kit (stem cell factor receptor), and are Ter119 (cell surface molecule)-positive. Their proliferative capacity is more limited compared to the preceding stage, the CFU-e.
In vivo, starting with the proerythroblast stage, erythroid cells undergo several more cell divisions while at the same time upregulating survival genes such as Bcl-xL, acquiring and storing large amounts of iron, ramping up the synthesis of hemoglobin and other erythroid genes (in large part a GATA-1 dependent process that is augmented by the EpoR signaling) and decreasing in cell size, eventually removing their nuclei and being released into the bloodstream as a reticulocyte.

Plasma cells (or plasmocytes) are cells of the immune system that secrete large amounts of antibodies. They differentiate from B cells upon stimulation by antigen-activated CD4+ lymphocytes (or T lymphocytes) . They are found in the medulla of the lymph node, in the bone marrow and in the red pulp of the spleen. The differentiation of B cells into plasma cells is accompanied by many morphological changes that allow the cell to synthesize and secrete large amounts of antibodies.

Erythropoiesis is the process by which red blood cells (erythrocytes) are produced in bone marrow. It is stimulated by decreased O2 in circulation, leading to secretion of the hormone erythropoietin by the kidneys. This hormone stimulates proliferation and differentiation of red cell precursors, which activates increased erythropoiesis in the hemopoietic tissues and ultimately producing red blood cells. In certain diseases, erythropoiesis also occurs outside the bone marrow, e.g. within the spleen or liver, and is known as extramedullary erythropoiesis. Individuals with anemia have abnormally low levels of red blood cells whereas those with polycythemia have abnormally high levels of red blood cells.

Megakaryopoiesis is the process by which bone marrow progenitor cells (hematopoietic stem cells, HSC) develop into mature megakaryocytes (MKs). MKs then produce platelets, a process called thrombopoesis. MKs play a role in the pathogenesis of acute megakaryocytic leukaemia and a subset of myeloproliferative neoplasms, including essential thrombocythemia and primary myelofibrosis. Abnormal megakaryopoiesis and thrombopoiesis can lead to morbidity and mortality from bleeding due to moderate to severe thrombocytopenia.
