They are unicellular prokaryotic microorganisms which generally possess rigid cell walls and multiply by cell division. They exhibit three principal forms: round or coccal, rodlike or bacillary, and spiral or spirochetal. Bacteria can be classified by their response to oxygen (aerobic, anaerobic, or facultatively anaerobic) and by how they obtain their energy (chemotrophic, i.e. via a chemical reaction or phototrophic, via a light reaction). In addition, chemotrophs obtain their energy from chemicals, lithotrophic from inorganic compounds and organotrophic from organic compounds. Bacteria are also classified by the source carbon that they utilize: heterotrophic, from organic sources or autotrophic, from carbon dioxide. They can also be classified by whether or not they stain (based on the structure of their cell walls) with crystal violet dye: gram-negative or gram-positive.

Trichomonas vaginalis (TV) is an anaerobic, flagellated parasitic protozoan that is among the most common non-viral sexually transmitted organisms whose only known host is humans. It is estimated that 170 million new cases occur worldwide and it is the most common sexually transmitted diseases in the United States. TV is known to increase HIV acquisition and transmission in both men and women by acting as a cofactor to increase susceptibility to HIV of an uninfected partner and infectivity of HIV of an infected partner. In women, infection with TV is a possible risk factor for developing cervical neoplasia, post-operative infections, adverse pregnancy outcomes, atypical pelvic inflammatory disease and infertility. In men, infection with TV can lead to non-gonococcal urethritis, infertility, chronic prostatitis and decreased sperm motility. Diagnosis is most often made using wet mount microscopy which has a sensitivity of only 52%. Other diagnostic methods include Pap smear, culture and PCR with sensitivities of 56%, 78% and 84%, respectively. There is an estimated 5 million new cases of TV annually in the United States and yet TV is still not a reportable disease. Treatment for TV is with metronidazole though emerging metronidazole-resistant strains have been reported. Asymptomatic infection can persist for months to years. Fungi are eukaryotic microorganisms which manifest as yeasts, molds or a combination. Yeasts start as individual cells that reproduce by budding. Molds, which are long filaments or hyphae, grow by apical extension. Both yeasts and molds are heterotrophic and digest their food externally by absorptive nutrition. Taxonomically, yeast are characterized by their size and shape, the presence or absence of capsules, conidiogenesis (the process of daughter cell formation), presence of sexual spores, the formation of pseudo-and true hyphae and physiologic data. Species are differentiated based on the ability to assimilate and ferment carbon sources, and to utilize nitrate as nitrogen. Some opportunistic fungal pathogens found in humans are dimorphic. They grow as mycelium in nature and reproduce as yeast in the body. Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. Erythrocytes or red blood cells (RBCs) are the cells in the circulation that carry oxygen to and remove carbon dioxide from the tissues throughout the body. They are produced in the bone marrow in response to erythropoietin where they transition through six stages over a seven day period. When they are released into the circulation, their nucleus has been extruded and they measure 6-8 microns in diameter. The lifespan of RBCs is about 120 days. When the RBC ages the cell membrane becomes less pliable and tears as they cell travels through the micro vessels of the body. The damaged RBCs are removed from the circulation by the spleen. Variations in number, shape and size of RBCs are indicative of many diseases and disorders. There are many factors that may lead to decreased numbers of RBCs including hemorrhage, hemolysis, iron or vitamin deficiency, marrow failure and more. A wet preparation or wet mount is used to identify the cause of vaginosis, a general term for infections of the vagina. A drop of vaginal discharge is placed on a slide with a drop of liquid -usually water--which is then examined microscopically as an unstained wet mount or as a Gram-stained slide. The refractive index of the water improves the image quality. Wet mounts generally cannot be stored since the water evaporates. Glycerol or immersion oil can be used instead of water.
