LP14304-7 Erythrocytes
Erythrocytes or red blood cells (RBCs) are the cells in the circulation that carry oxygen to and remove carbon dioxide from the tissues throughout the body. They are produced in the bone marrow in response to erythropoietin where they transition through six stages over a seven day period. When they are released into the circulation, their nucleus has been extruded and they measure 6-8 microns in diameter. The lifespan of RBCs is about 120 days. When the RBC ages the cell membrane becomes less pliable and tears as they cell travels through the micro vessels of the body. The damaged RBCs are removed from the circulation by the spleen. Variations in number, shape and size of RBCs are indicative of many diseases and disorders. There are many factors that may lead to decreased numbers of RBCs including hemorrhage, hemolysis, iron or vitamin deficiency, marrow failure and more. Larger than normal RBCs may be indicative of liver disease while smaller than normal RBCs are seen in thalassemias and other anemias.(Mosby's manual of diagnostic and laboratory tests, Kathleen Deska Pagana; Timothy James Pagana, Elsevier St. Louis, Mo ©2010)

LP14419-3 Leukocytes
Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. LP31538-9 Leukocyte clumps
Leukocyte clumps are white blood cells grouped together by agglutination. Leukocyte clumps in urine indicate the presence of an infection or inflammation, especially in the kidney (e.g. pyelonephritis, interstitial inflammation, parenchymal infections). Leukocyte clumps in the blood may be associated with cancer, including non-Hodgkins lymphoma and chronic lymphocytic leukemia.

LP70260-2 Epithelial cells.squamous
Epithelial cells cover the inside and outside of almost all of the body's surfaces. Squamous epithelial cells are flattened and barely visible by light microscopy. They fit closely together in tissues; providing a smooth, low-friction surface over which fluids can move easily. Squamous epithelia are found lining surfaces such as the alveolar epithelium in the lungs. Specialized squamous epithelia also form the lining of cavities such as the blood vessels (endothelium) and pericardium (mesothelium) and the major cavities found within the body.

LP14082-9 Bacteria
One of the three domains of life (the others being Eukarya and Archaea), also called Eubacteria. They are unicellular prokaryotic microorganisms which generally possess rigid cell walls and multiply by cell division. They exhibit three principal forms: round or coccal, rodlike or bacillary, and spiral or spirochetal. Bacteria can be classified by their response to oxygen (aerobic, anaerobic, or facultatively anaerobic) and by how they obtain their energy (chemotrophic, i.e. via a chemical reaction or phototrophic, via a light reaction). In addition, chemotrophs obtain their energy from chemicals, lithotrophic from inorganic compounds and organotrophic from organic compounds. Bacteria are also classified by the source carbon that they utilize: heterotrophic, from organic sources or autotrophic, from carbon dioxide. They can also be classified by whether or not they stain (based on the structure of their cell walls) with crystal violet dye: gram-negative or gram-positive.

LP16849-9 Crystals
Crystals are formed in urine when solutes precipitate out of solution. Crystals are typically absent in freshly voided urine. Normally they precipitate as the urine cools or after it is refrigerated. Most crystals are not indicative of a pathological process, but when present in freshly voided urine, they indicate that crystals are forming within the body which is clinically important. When crystals develop within the nephrons, serious tubular damage is possible. Since the presence of some crystals in the urine are considered pathological, it is crucial that they be correctly identified and reported. Crystal identification is made based on microscopic physical appearance and urine pH.[Nancy A. Brunzel, Fundamentals of Urine & Body Fluid Analysis, p 189]

Part Descriptions
LP16857-2 Hyaline casts
The most common type of casts, hyaline casts are solidified Tamm-Horsfall mucoproteins secreted from the tubular epithelial cells of individual nephrons. Low urine flow, concentrated urine, or an acidic environment can contribute to the formation of hyaline casts, and, as such, they may be seen in normal individuals in dehydration or vigorous exercise. Hyaline casts are cylindrical and clear, with a low refractive index, so that they can easily be missed on cursory review under brightfield microscopy, or in an aged sample where dissolution has occurred. On the other hand, phase contrast microscopy leads to easier identification. Given the ubiquitous presence of Tamm-Horsfall protein, other cast types are formed via the inclusion or adhesion of other elements to the hyaline base.

LP14044-9 Casts
Urinary casts are cylindrical aggregations of particles that form in the distal nephron of the kidney and can be seen in urine samples examined by light microscopy. Many types of casts exist, including red blood cell, white blood cell, and fatty casts. Mixed cellular casts are casts that contain multiple types of cells. Different types of casts are associated with different diseases, for example, white blood cell casts are indicators of kidney infection, and fatty casts can be seen in patients with nephrotic syndrome.[MedlinePlus Encyclopedia: 003586]

LP14765-9 Spermatozoa
A spermatozoon or spermatozoan (pl. spermatozoa), from the ancient Greek (seed) and (living being) is the haploid cell that is the male gamete. After a female ovum is fertilized by a sperm cell it becomes a zygote. Sperm cells contribute half of the genetic information to the diploid offspring and the other hallf is contributed by the female ovum. In mammals, the sex of the offspring is determined by the sperm cells: a spermatozoon bearing a Y chromosome will lead to a male (XY) offspring, whereas one bearing an X chromosome will lead to a female (XX) offspring (the ovum always provides an X chromosome). Sperm cells were first observed by a student of Antoni van Leeuwenhoek in 1677.
