Glucan 1,4-alpha-glucosidase (EC 3.2.1.3), also known as glucoamylase, is part of the maltase-glucoamylase (MGA) complex found in the mucosal brush border of the small intestine. MGA plays a role in starch digestion, and more specifically, glucan 1,4-alpha-glucosidase successively hydrolyzes terminal-linked alpha-D-glucose residues from the non-reducing ends with release of beta-D-glucose. Deficiency of intestinal glucan 1,4-alpha-glucosidase is associated with chronic diarrhea that responds to dietary starch elimination. PMID: 9446624

Lactase (EC 3.2.1.108) one part of the β-galactosidase family of enzymes, is a glycoside hydrolase involved in the hydrolysis of the disaccharide lactose into constituent galactose and glucose monomers. (http://enzyme.expasy.org/EC/3.2.1.108) Lactase is present predominantly along the brush border membrane of the differentiated enterocytes lining the villi of the small intestine. In humans, lactase is encoded by the LCT gene.

Lactose is a sugar found in milk and milk products. The small intestine, the organ where most food digestion and nutrient absorption take place, produces an enzyme called lactase, which breaks down lactose into two simpler forms of sugar: glucose and galactose. People with small intestinal lactase deficiency cannot digest much lactose, which may cause lactose malabsorption. In lactose malabsorption, undigested lactose passes to the colon, where bacteria break down undigested lactose and create fluid and gas. Not all people with lactase deficiency and lactose malabsorption have digestive symptoms. Lactose intolerance is usually diagnosed by patient history, physical exam, and the hydrogen breath test and/or stool acidity test. Although it is rarely done, the reference standard for diagnosing lactase insufficiency is directly measuring lactase activity in a duodenal biopsy specimen.

Sucrase is the name given to a number of enzymes that catalyse the hydrolysis of sucrose to fructose and glucose. The enzyme invertase, which occurs more commonly in plants, also hydrolyzes sucrose but by a different mechanism.
Sucrose intolerance (also known as Congenital Sucrase-Isomaltase Deficiency (CSID) or Sucrase-isomaltase deficiency) occurs when sucrase is not secreted in the small intestine. With sucrose intolerance, the result of consuming sucrose is excess gas production and often diarrhea and malabsorption. Lactose intolerance is a related disorder that reflects an individual's inability to hydrolyze the disaccharide lactose.
Sucrase is secreted by the tips of the villi of the epithelium in the small intestine. Its levels are reduced in response to villi-blunting events such as celiac sprue and the inflammation associated with the disorder. The levels increase in Pregnancy/Lactation and Diabetes as the villi hypertrophy.
