Von Willebrand factor (vWF) is a high-molecular-weight plasma protein produced by endothelial cells and megakaryocytes. It is part of the factor VIII/vWF complex. The vWF has receptors for collagen, platelets, and ristocetin. Ristocetin is an antibiotic, previously used to treat staphylococcal infections, now mainly used in vitro to diagnose conditions such as von Willebrand disease. Platelet agglutination caused by ristocetin can occur only in the presence of von Willebrand factor multimers, so if ristocetin is added to blood lacking the factor it will not coagulate. The vWF facilitates the adhesion of platelets to collagen and hemostatic plug formation. The prolonged bleeding time in von Willebrand's disease is due to the deficiency of this factor.
