Glimepiride is a medium-to-long acting sulfonylurea anti-diabetic drug. It is marketed as Amaryl and Glista OD. Glimepiride is the first third-generation sulfonylurea, and is very potent. It is used to treat type 2 diabetes mellitus by increasing insulin production in the pancreas. Side effects include gastrointestinal tract (GI) disturbance, and rarely thrombocytopenia, leukopenia, hemolytic anemia, and allergic reactions occur.

Repaglinide (Prandin in the U.S., GlucoNorm in Canada, NovoNorm elsewhere) is used for the treatment of type II diabetes. It lowers blood glucose by stimulating the release of insulin from the pancreas. It achieves this by closing ATP-dependent potassium channels in the membrane of the beta cells. This depolarizes the beta cells, opening the cells' calcium channels, and the resulting calcium influx induces insulin secretion. The half life of Repaglinide is 1 hour.

LP16296-3 TOLBUTamide
Tolbutamide is a first generation sulfonylurea oral hypoglycemic drug sold under the brand name Orinase. This drug may be used in the management of type II diabetes if diet alone is not effective. Tolbutamide stimulates the secretion of insulin by the pancreas. Since the pancreas must synthesize insulin in order for this drug to work, it is not effective in the management of type I diabetes. It is not routinely used due to a higher incidence of adverse effects compared to newer second generation sulfonylureas, such as glyburide.

Sulfonylurea (UK: Sulphonylurea) derivatives are antidiabetic drugs primarily used to treat diabetes mellitus type 2, and include drugs such as Chlorpropamide, Tolazamide, Tolbutamide, Glyburide, Gliclazide, Glipizide, Gliquidone, Acetohexamide and others. They act by increasing insulin release from the beta cells in the pancreas. Sulfonylureas are ineffective where there is absolute deficiency of insulin production such as in type 1 diabetes or post-pancreatectomy.
