Prostate cancer gene 3 (PCA3) is strongly expressed in 95% of primary prostate cancer specimens and in prostate cancer metastasis. PCA3 noninvasive molecular amplification assay detects and quantifies both PCA3 and PSA mRNA levels, providing a PCA3/PSA ratio score that can assist in determining whether a patient is at increased risk for developing prostate cancer. Patients positive for PCA3 have PCA3 scores greater than 35 and are at increased risk for prostate cancer.

PCA3 and PSA mRNA are isolated from urine specimens and amplified by transcription mediated amplification (TMA). Detection is achieved by hybridization protection assay (HPA). PCA3 and PSA mRNAs are quantified and the PCA3 Score is determined (PCA3 Score = [PCA3 mRNA]/[PSA mRNA] x1000). Source: LOINC partner 1
