Glucose 6-Phosphate Dehydrogenase (G6PD) deficiency causes hemolytic anemia following the use of some drugs, bacterial or viral infections, or metabolic disorder such as acidosis. Three G6PD variants occur frequently and have clinical implications in different ethnic groups. Heinz bodies may appear in peripheral red blood cells when G6PD-mediated hemolysis occurs. Younger red cells and reticulocytes contain more G6PD and may cause normal values if blood is collected following a hemolytic crisis.

ATP:pyruvate 2-O-phosphotransferase. A phosphotransferase that catalyzes reversibly the phosphorylation of pyruvate to phosphoenolpyruvate in the presence of ATP. It has four isozymes (L, R, M1, and M2). Deficiency of the enzyme results in hemolytic anemia. EC 2.7.1.40.

Pyruvate Kinase deficiency, although rare, is the erythrocyte enzyme deficiency most frequently found to be a cause of congenital nonspherocytic hemolytic anemia. Patients usually present early in childhood with anemia, icterus, and splenomegaly. Hemolytic disease of the newborn is common in persons with PK deficiency.

Phosphofructokinase (PFK) is a kinase enzyme that phosphorylates fructose-6-phosphate to fructose-1,6-bisphosphate in the glycolytic pathway. PFK exists as a homotetramer in bacteria and mammals (where each monomer possesses 2 similar domains) and as an octomer in yeast (where there are 4 alpha- (PFK1) and 4 beta-chains (PFK2)). Deficiency in PFK leads to glycogenosis type VII (Tarui's disease), an autosomal recessive disorder characterised by severe nausea, vomiting, muscle cramps and myoglobinuria in response to bursts of intense or vigorous exercise. Individuals with PFK deficiency are usually able to relieve symptoms by adjusting activity levels.

Adenosine deaminase (ADA) is a purine salvage pathway enzyme, which catalyzes the irreversible deamination of adenosine into inosine and ammonia. Elevated adenosine deaminase activities are found in tuberculous pleural and pericardial effusions. In addition, increased ADA levels in cerebrospinal fluid (CSF) have been observed in tuberculous meningitis. These ADA increases are caused by the cellular immune response in the fluid compartment, especially in the activation of T lymphocytes. Thus, many disorders can cause elevations. Deficency of adenosine deaminase is the most frequent known cause of SCID (Severe combined immunodeficency disease) it is observed in 25% of cases.

Aldolase is an enzyme which catalyzes the aldol reaction. It beaks down fructose 1,6-bisphosphate (F-1,6-BP) into glyceraldehyde 3-phosphate and dihydroxyacetone phosphate (DHAP), as part of glycolysis. In mammalian aldolase the key catalytic amino acid residues involved in the reaction are lysine and tyrosine. Tyrosine acts as an efficient hydrogen acceptor whereas the lysine covalently binds and stabilized the intermediates. Many bacteria use two magnesium ions in place of the lysine.

Aldolase glycolytic enzyme that splits fructose 1,6-diphosphate. It is present most significantly in skeletal and heart muscle, although all cells contains samll amount, and the liver has a moderate concentration. Damage to skeletal muscle produces high levels of aldolase; this is particularly so in the case of progressive muscular dystrophy in which levels may be 10-15 times normal. There is no aldolase elevation in muscle diseases of neural origin such as neurogenic muscular atrophy, poliomyelitis, myasthenia gravis and multiple sclerosis, but any of the inflammatory conditions involving muscle may cause moderately elevated values. Used for the differential diagnosis of Duchenne's muscular dystrophy and in carriers lesser degrees of elevation in dermatomyositis, polynyositis, and limb-girdle dystrophy. In recent years, CK is used more often for diagnosis of muscular dystrophy.

Triose-phosphate isomerase (TPI or TIM), is an enzyme (EC 5.3.1.1) that catalyzes the reversible interconversion of the triose phosphate isomers dihydroxyacetone phosphate and D-glyceraldehyde 3-phosphate.
In humans, deficiencies in TPI are associated with a progressive, severe neurological disorder called triose phosphate isomerase deficiency. Triose phosphate isomerase deficiency is characterized by chronic hemolytic anemia. While there are various mutations that cause this disease, most include the mutation of glutamic acid at position 104 to aspartic acid.

Bisphosphoglycerate mutase (BPGM) is an enzyme unique to erythrocytes and placental cells. BPGM is responsible for the catalytic synthesis of 2,3-Bisphosphoglycerate (2,3-BPG) from 1,3-BPG. Bisphosphoglycerate has a mutase and a phosphatase function, but these are much less active, in contrast to its glycolitic cousin, phosphoglycerate mutase (PGM), which favors these two functions, but can also catalyze the synthesis of 2,3-BPG to a lesser extent. Deficiencies of BPGM and bisphosphoglycerate phosphatase (BPGP) are associated with mild erythrocytosis due to absence of 2,3-diphosphoglycerate (2,3-DPG).
