Lactase (EC 3.2.1.108) one part of the β-galactosidase family of enzymes, is a glycoside hydrolase involved in the hydrolysis of the disaccharide lactose into constituent galactose and glucose monomers. (http://enzyme.expasy.org/EC/3.2.1.108) Lactase is present predominantly along the brush border membrane of the differentiated enterocytes lining the villi of the small intestine. In humans, lactase is encoded by the LCT gene.

Lactose is a sugar found in milk and milk products. The small intestine, the organ where most food digestion and nutrient absorption take place, produces an enzyme called lactase, which breaks down lactose into two simpler forms of sugar: glucose and galactose. People with small intestinal lactase deficiency cannot digest much lactose, which may cause lactose malabsorption. In lactose malabsorption, undigested lactose passes to the colon, where bacteria break down undigested lactose and create fluid and gas. Not all people with lactase deficiency and lactose malabsorption have digestive symptoms. Lactose intolerance is usually diagnosed by patient history, physical exam, and the hydrogen breath test and/or stool acidity test. Although it is rarely done, the reference standard for diagnosing lactase insufficiency is directly measuring lactase activity in a duodenal biopsy specimen.
