Programmed cell death-ligand 1 (PD-L1), also known as cluster of differentiation 274 (CD274) or B7 homolog 1 (B7-H1), is a protein that in humans is encoded by the CD274 gene. PD-L1 is the major ligand for the programmed cell death-1 (PD-1) receptor, which is present on the surface of T, B and NK cells and is part of the B7-CD28 superfamily. Binding PD-L1 and PD-L2 to the PD-1 receptor found on T-cells inhibits T cell proliferation and cytokine production. An increase PD-L1 expression occurs in some tumors, which helps protect them against active T-cell immune surveillance by binding to PD-1 and inhibiting the T-cell response. Positive PD-L1 immunohistochemistry in tumor cells may predict tumor response to therapy with immune checkpoint inhibitors, which are a new class of drugs that trigger an immune response against cancer cells by effectively suppressing the tumor's ability to inhibit the immune response. PMID: 25682184 Drugs in this class include pembrolizumab and nivolumab, both of which are anti-PD-1 receptor antibodies, and atezolizumab, which is an anti-PD-L1 antibody.
