72318-9 Centromere protein A IgG Ab [Presence] in Serum or Plasma by Immunoassay
Term Description
Centromere protein A (CENP-A) IgG class autoantibodies can be detected by enzyme immunoassays. Results may be interpreted by humans or automated methods and used for the diagnosis of system sclerosis (SSc, diffuse and limited forms) and overlapping syndromes. This code was created for, but not limited to, the detection of CENP-A autoantibodies in Euroimmun's Systemic Sclerosis (Nucleoli) Profile EUROLINE (IgG) test kit.

Part Descriptions
LP157222-3 Centromere protein A
Centromere protein A (CENP-A) is an essential centromere protein encoded by the CENPA gene. Overexpression of CENP-A causes chromosomal aberrations, and depletion of CENP-A promotes apoptosis and induces cell cycle arrest. Overexpression of CENP-A has been associated with high grade cancers and is a prognostic factor for patients with estrogen receptor (ER)-positive breast cancer. Antibodies to CENP-A are associated with scleroderma, systemic sclerosis, CREST syndrome, and other rheumatologic diseases.

- The PrThr property is used for LOINC terms whose results are reported using an ordered categorical scale, regardless of whether or not an internal threshold was used to make that determination. This change was approved by the Laboratory LOINC Committee in June 2016.; The EIA method, which was always intended to cover more than just enzyme-linked immunoassay and whose display name for the long common name has always been Immunoassay, was renamed IA in order to eliminate ambiguity about whether the method has a broader meaning than just enzyme-linked immunoassay. Likewise, EIA.rapid was renamed IA.rapid. These changes were approved by the Laboratory LOINC Committee in June 2016.

82913-5 Centromere protein B IgG Ab [Presence] in Serum by Line blot
Term Description
This term was created for, but is not limited in use to, the submitter's test strip, a membrane strip pre-coated with a specific set of antigens in parallel lines. If antibodies to any of those antigens are present in the patient's serum, they will bind to the specific antigen. The bound patient antibodies are then detected using labelled anti-human antibodies. Various manufacturers classify this method as an immunoblot technique. LOINC uses the immunoblot (IB) method to mean that some electrophoretic/chromatographic separation occurs such that position (size) of the band is part of the assessment (as in Western blot). Because this assay does not involve a separation step but rather uses strips with pre-fixed antigens, the LOINC Method is Line blot.

- This term was created for an assay that uses a membrane strip pre-coated with a specific set of antigens in parallel lines. Because the antigens are seperated by electrophoresis/chromatographic separation during manufacturing, not during the test procedure itself, this test was assigned a LOINC Method of Immunoassay (IA) and not Immunoblot (IB). However, upon further review of IA, IB, and Immunoblot strip methodologies and regulatory distinctions between these, we changed the Method from IA to Line blot so that users can distinguish between these three Methods. The meaning of the term has not changed since it was created for an IB strip assay as described in the term description.

85095-8 D-3-phosphoglycerate dehydrogenase IgG Ab [Presence] in Serum by Line blot
Term Description
The qualitative result for the detection of IgG antibodies to D-3-phosphoglycerate dehydrogenase (PGDH) antigens in serum or plasma for the diagnosis of autoimmune liver diseases, including autoimmune hepatitis and primary biliary cirrhosis. This term was created for, but is not limited in use to, the submitter's test strip, a membrane strip pre-coated with a specific set of antigens in parallel lines. Part Descriptions
LP200136-2 D-3-phosphoglycerate dehydrogenase
D-3-phosphoglycerate dehydrogenase (3-PGDH) is encoded by the PHGDH gene, located on chromosome 1p12, and functions as part of the L-serine biosynthesis pathway to catalyze the conversion of 3-phospho-D-glycerate to 3-phosphohydroxypyruvate. Defects in the PHGDH gene are associated with two disorders: phosphoglycerate dehydrogenase(PGDH) deficiency and Neu-Laxova syndrome 1 (NLS1). PGDH deficiency is associated with congenital microcephaly, psychomotor retardation, and seizures. NLS1 is a lethal, autosomal recessive syndrome associated with ichthyosis, marked intrauterine growth restriction, microcephaly, hypoplastic lungs and central nervous system anomalies. [UniProt: O43175] Antibodies to 3-PGDH are detected in patients with chronic liver diseases, primiarily autoimmune hepatitis (AIH). PMID: 21707887

LP157237-1 Mitochondria M2-3E
M2-3E (BPO) antigen is a recombinant fusion protein that is made up of the E2 subunits of the branched-chain 2-oxoacid dehydrogenase (BCOADH), pyruvate dehydrogenase (PDH) and 2-oxoglutarate dehydrogenase (OGDH). The presence of anti-mitochondrial antibodies (AMA) against M2-3E antigens is used for the diagnosis of primary biliary liver cirrhosis (PBC), an immune-mediated chronic inflammatory liver disease of unknown cause. Approximately 86% of patients with PBC have M2-3E autoantibodies.

82931-7 Nuclear pore protein gp210 IgG Ab [Presence] in Serum by Line blot

LP71756-8 Nuclear pore protein gp210
Nuclear pore glycoprotein-210 (gp210) is an essential trafficking regulator in the eukaryotic nuclear pore complex. Gp-210 anchors the pore complex to the nuclear membrane and protein tagging reveals its primarily located on the luminal side of double layer membrane at the pore. A single polypeptide motif of gp210 is responsible for sorting to nuclear membrane, and indicate the carboxyl tail of the protein is oriented toward the cytoplasmic side of the membrane.

Part Descriptions
LP16758-2 SCL-70 extractable nuclear
Topoisomerase I (also known as Scl-70) is a nuclear enzyme that plays a role in releasing tension that is introduced in double-stranded DNA during DNA replication by transiently cleaving and rejoining one strand of the DNA duplex. Anti-Scl-70 autoantibodies are a marker of systemic sclerosis as well as other autoimmune diseases, including systemic lupus erythematosus (SLE). In patients with systemic sclerosis, anti-SCL-70 may be associated with more severe disease, and in patients with SLE, anti-SCL-70 suggests an increased risk of pulmonary hypertension and renal involvement. In general, anti-SCL-70 antibody status may help with diagnosis, treatment planning and prognosis in patients with connective tissue disease. PMID: 11229469

LP16760-8 Sjogrens syndrome-A extractable nuclear
Sjogren syndrome type A antigen (also known as SS-A, Ro, and E3 ubiquitin-protein ligase TRIM21) and Sjogren syndrome type B antigen (also known as SS-B and Lupus La protein) are nuclear antigens to which autoantibodies develop in autoimmune disorders such as Sjogren's syndrome (SS) and systemic lupus erythematosus (SLE). Between 40 and 70% of patients with SS have anti-SSA and/or anti-SSB autoantibodies, and 30-40% and 10-15% of patients with SLE are anti-SSA and anti-SSB positive, respectively. Longitudinal studies have shown that anti-SSA and -SSB antibodies may be present up to 20 years before the clinical onset of disease. SS patients who are anti-SSA and anti-SSB positive may have longer disease duration, parotid gland enlargement, decreased salivary flow, more severe minor salivary gland infiltration and a higher prevalence of extraglandular manifestations.

- Change Reason
