The deepest extent or maximum degree of local tumor invasion into or through the bowel wall. Results are based on the T component of the TNM staging system, as outlined in the AJCC Cancer Staging Manual. PMID: 20180029 Invasion depths include submucosa (pT1), muscularis propria (pT2), through muscularis propria into pericolorectal tissues (pT3), penetrating the visceral peritoneum (pT4a), and directly invading or adherent to other organs or structures (pT4b). This term was created for, but not limited in use to, the Colorectal Cancer Structured Reporting Protocol (2nd Edition, 2012) developed by the Royal College of Pathologists of Australasia (RCPA).

The result for whether or not the circumferential resection margin (CRM) on the non-peritonealized surface of the resected specimen is involved. CRM involvement (1mm or less distance between tumor and CRM) is an important predictor of local recurrence in rectal carcinoma. Biopsies of the skin are done in order to remove skin lesions and determine the specific diagnosis. Surgical margins are the boundaries of the excised specimen, and in the case of a skin biopsy, the deep margin is the one furthest away from the skin surface. A margin is considered 'positive' if it contains neoplastic cells and indicates that additional treatment may be needed besides the excision. Deeper excisions are more invasive but are associated with diminished chances for the tumor's reoccurrence. PMID: 25484385

Microsatellite instability (MSI) is a change in the number of repeats of a specific DNA marker in cancer cells compared to the number of repeats present in non-cancerous cells of the same patient. In cancer, aberrant DNA methylation patterns are found in two distinct forms: hypermethylation and hypomethylation. Hypermethylation is one of the major epigenetic modifications that repress transcription via the promoter region of tumor suppressor genes. Hypermethylation typically occurs at CpG islands in the promoter region and is associated with gene inactivation. Hypomethylation, linked to chromosomal instability and loss of imprinting, arises earlier and occurs throughout the progression of cancer.
