Detection of FOXO1 rearrangements with several translocation partners, including PAX3 and PAX7, by FISH analysis. FISH studies are shown to detect rearrangements not found by PCR, and they will often yield results when the quality of the specimen is poor or the PCR results are equivocal.

The FOXO1 gene is part of the forkhead family of transcription factors but its specific function is not yet known. Gene rearrangements involving the FOXO1 are seen frequently in alveolar rhabdomyosarcoma (ARMS), an aggressive soft-tissue sarcoma. PMID: 19704258

FISH (fluorescence in situ hybridization) is a cytogenetic technique used to detect and localize the presence or absence of specific DNA sequences on chromosomes. FISH uses fluorescent probes that bind to only those parts of the chromosome with which they show a high degree of sequence similarity. Fluorescence microscopy can be used to find out where the fluorescent probe bound to the chromosomes. FISH is often used for finding specific features in DNA for use in genetic counseling, medicine, and species identification.
