Basophils are the least common of the granulocytes, representing about 0.5% to 1% of circulating white blood cells. They contain large cytoplasmic granules which partially hide the cell nucleus when examined under the microscope. However, when unstained, the nucleus is visible and it usually has 2 lobes. A cell in tissues, the mast cell, has many similar characteristics. For example, both cell types store histamine, a chemical that is secreted by the cells when stimulated in certain ways (histamine causes some of the symptoms of an allergic reaction). Like all circulating granulocytes, basophils can be recruited out of the blood into a tissue when needed. When activated, basophils secrete histamine, several proteoglycans, lipid mediators like leukotrienes, and several cytokines. Histamine and proteoglycans are pre-stored in the cell's granules whereas the other secreted substances are newly synthesized. Each of these substances contributes to inflammation. Basophils are granular white blood cells characterized by a pale-staining nucleus with 2 to 3 lobes and a cytoplasm containing coarse, dark-staining granules of variable size. They represent 0.5-1% of circulating leukocytes and they are stained by basic dyes. When activated, they release histamine, leukotrienes and a number of cytokines.
