Soluble C3-convertase, also known as iC3Bb, catalyzes the proteolytic cleavage of complement component C3 into C3a and C3b during the alternate complement pathway. C3b binds to microbial pathgens leading to the production of surface-bound C3 convertase and then to more C3b. Also known as C3bBb, this convertase is similar to soluble C3-convertase except that it is membrane bound. Bound C3b facilitates opsonization of pathogens by macrophages. A glycoprotein central in both the classical and alternate pathways of complement systems. C3 can be cleaved into C3a and C3b fragments either spontaneously at low level, or at a higher rate by C3 convertase. C3a is an anaphylotoxin and a mediator of local inflammatory process. The complement system is a key part of the immune system and inflammatory response. The system has three pathways: 1) the classic pathway, 2) the alternative pathway, and 3) the lectin activation pathway. Complement C3 is involved in the activation of both the classical and alternate complement pathways. It is the most concentrated of the complement members and is composed of 5 subunits: C3a, C3b, C3c, C3d and C3 d. Decreased levels of C3 is associated with disease inactivation in systemic lupus erythematosus (SLE) and may also be associated with acute glomerulonephritis, immune complex disease, septic shock, and end-stage liver disease.
