Platelet-derived growth factor receptors (PDGFRs) are cell surface tyrosine kinase receptors for members of the platelet-derived growth factor (PDGF) family. PDGF subunits -A and -B are important factors regulating cell proliferation, cellular differentiation, cell growth, development and many diseases including cancer. There are two forms of the PDGFR-- alpha and beta -- each encoded by a different gene. Depending on which growth factor is bound, PDGFR homo- or heterodimerizes. Like many other growth factors that have been linked to disease, PDGF and its receptors have provided a market for receptor antagonists to treat disease. Such antagonists include (but are not limited to) specific antibodies that target the molecule of interest, which act only in a neutralizing manner. Platelet-derived growth factor receptors (PDGFRs) exhibit tyrosine-protein kinase activity and have been implicated in the control of cell proliferation, survival and migration. PDGF receptors, PDGFR-alpha and PDGFR-beta, have 5 extracellular immunoglobulin-like domains and an intracellular tyrosine kinase domain. Upon binding a PDGF, the receptors form homo-and heterodimers. Dimerization of the receptors results in phosphorylation in the complex. More than 10 different SH2-domain-containing molecules have been shown to bind to different autophosphorylation sites in the PDGF-alpha and -beta receptors. PDGF-alpha receptors are expressed in oligodendrocyte progenitor cells and PDGF beta receptors are expressed on neurons. Source: LOINC partner 1
