Description

Daclatasvir (also called DCV, formerly called BMS-790052, tradename Daklinza) is a first-generation, direct-acting antiviral (DAA) is a zinc-binding and proline-rich hydrophilic phosphoprotein that plays a key role in hepatitis C virus RNA replication. Daclatasvir is used along with other medications, such as sofosbuvir (tradename Solvadi), to treat certain types of chronic hepatitis C. Treatment stops the virus from multiplying and in turn prevents secondary diseases of hepatitis C, including cirrhosis and liver cancer. Patients who have no detectable virus in the blood for a longer a period following treatment may be at lower risk of such secondary diseases. Six different forms of hepatitis C viruses are now known, referred to as genotypes 1 to 6. In clinical studies, the baseline NS5A polymorphism Y93H resulted in reduced rates of sustained virologic response in patients with HCV genotype 3 who were treated for 12 weeks with daclatasvir (plus sofosbuvir), especially in those patients with cirrhosis. In addition, the high mutation rate of the HCV genome, combined with selective pressure from ongoing therapy, can lead to selection of additional HCV variants that are resistant to NS5A inhibitors.
