Description
The activated partial thromboplastin time (aPTT) is used to evaluate the functionality of the intrinsic pathway of coagulation. The aPTT can only detect deficiencies of factors VIII, IX, XI, and XII and only if the deficiency is 20-40% of normal. PMID: 23718922 The aPTT clotting time can vary depending upon a number of factors including its activator composition, the platelet phospholipid substitute, the calcium chloride concentration, and incubation time. All of these various properties also affect how sensitive the aPTT is at detecting clotting factor deficiencies, either functional or quantitative. PMID: 3940420 Some aPTT reagents have been found to be more sensitive in being able to detect most of the factor VIII, IX, and XI deficiencies. An aPTT assay is run using patient plasma that is diluted with pooled normal plasma to determine if there is a factor deficiency or an inhibitor. PMID: 20840376 The detection limits of factor deficient plasma can be obtained from the reagent manufacturer for specific instruments. It is also possible to develop a detection limit by testing at least 20 factor deficient specimens that have activities that range from 10 to 50% for each of the intrinsic coagulation factors.
