Description
Rifampicin (RIF) is a first line drug used to treat Mycobacterium tuberculosis (TB). It inhibits transcription by binding with the rpoB encoded beta-subunit of the DNA-dependent RNA polymerase (RNAP). The majority of the mutations across bacterial species occurs in the 81 bp region of cluster, also known as the rifampicin resistance determining region (RRDR). The RRDR is surrounded by 12 amino acids, 11 of which have been linked to RIF mutations. The mutations in the RRDR usually result in an amino acid substitution that has an associated compact side chain with a larger amino acid and compact side chain. The inflexibility of rifampicin prevents it from being able to bind to the mutated beta subunit of RNAP. PMID: 26038512 Source: Regenstrief LOINC
