Description

Rearrangements or fusions involving ABL-class genes, including ABL1, ABL2 and PDGFRB, are associated with BCR-ABL1-like B-lymphoblastic leukemia/lymphoma (BCR-ABL1-like B-ALL), also known as Philadelphia-like (Ph-like) ALL. Ph-like ALL is characterized as a neoplasm of B lymphoblasts having gene expression similar to BCR-ABL1+ B-ALL cases but lacking the BCR-ABL1 translocation. Several methods are used to diagnose Ph-like ALL, including gene expression profiling (the "gold standard"), conventional karyotyping, fluorescence in situ hybridization (FISH), reverse-transcription polymerase chain reaction (RT-PCR), and various sequencing techniques. For positive FISH studies, additional fusion probe or RT-PCR testing is needed to identify translocation partners. Patients with Ph-like ALL have a poor prognosis but respond well to targeted therapy when the genetic cause is identified.
