Crystal violet stain
Active
Description
Crystal violet is a triphenylmethane dye most commonly used for gram-staining procedures and classifying bacteria. Gram positive bacteria stain crystal violet whereas gram negative cells do not retain the crystal violet and stain red. This staining method is also used to detect cytotoxicity or cell death causes by chemicals, drugs or toxins from pathogens. Crystal violet measures cell death since it binds to proteins and DNA. When cells undergo cell death, they lose their adherence to cell culture plates and are subsequently lost from the population of cells, reducing the amount of crystal violet staining in a culture.
