Cholecalciferol, also known as Vit D3, is a fat-soluble prohormone. Vitamin D has many functions including the regulation of calcium and phosphorus levels in blood, the promotion of bone formation and mineralization, and aid to the immune system by supporting phagocyctosis, anti-tumor activity, and immunomodulatory functions. A deficiency of vitamin D can be due to many causes including inadequate dietary intake, insufficient sunlight, malabsorption problems, inability to convert vitamin D into its active metabolite, kidney or liver impairment and many others. Vitamin D occurs naturally only in fish. It is produced in the skin when UVB radiation from sunlight reacts with 7-dehydrocholesterol (a derivative of cholesterol) to form pre-vitamin D3. The pre-vitamin D3 isomerizes to Vitamin D and is then hydroxylated in the liver to 25-hydroxycholecalciferol. 25-hydroxycholecalciferol is in turn hydroxylated in the kidney into two dihydroxylated metabolites which are 1,25-dihydroxycholecalciferol (the active hormone) and 24R,25 (OH)2D3.
