Amphetamine (AMPH) is a drug that stimulates both the central and peripheral nervous systems and is used to treat attention-deficit/hyperactivity disorder, narcolepsy and obesity. PMID: 17209801 AMPH increases extracellular monoamine neurotransmitters in the brain including dopamine, serotonin and norepinephrine. AMPH promotes presynaptic release of the neurotransmitters rather than blocking their re-uptake. Physiological responses to AMPH use include increased blood pressure, increased heart rate, increased body temperature, relaxed bronchial muscle and depressed appetite. Abuse of AMPH can cause powerful psychological dependence, tolerance, mild physical dependence associated with tachycardia, restlessness, irritability, insomnia, personality changes and an intense form of chronic intoxication psychosis similar to schizophrenia. Long-term users of AMPH can experience depression, impaired memory and motor skills most likely due to a drop in dopamine transporters and damage to dopaminergic and serotonergic neurons. Excretion of AMPH is in the urine and the rate of elimination is dependent on the pH of the urine. Urine with an alkaline pH has an excretion rate of 18-34 hours while that of acid urine is 7-14 hours.(Tietz Fundamentals of Clinical Chemistry and Molecular Diagnostics, Seventh Edition; Carl A.
