Hemoglobin S (HbS) is a variant form of hemoglobin found in people with sickle cell disease. The Hb S variant occurs primarily among people of African, Hispanic, and Mediterranean descent. Hb S has a variation in the β-chain gene that causes a change in the properties of hemoglobin, resulting in abnormal, rigid, sickling shape of red blood cells. Individuals with two copies of the Hb S variant have classic sickle cell disease (sickle cell anemia, HbSS). Individuals with one HbS variant and one normal hemoglobin, Hb A, have sickle cell trait (HbAS) and usually have no associated symptoms. Hemoglobins exhibit different migration patterns in alkaline versus acidic electrophoresis conditions. So a combination of 2 procedures can be used in tandem to identify the hemoglobin variants.
