U1 small nuclear ribonucleoprotein particle (U1 snRNP) is an RNA-protein complex that is composed of ten proteins: U1-70kD, U1A, U1C and seven Smith antigen (Sm) proteins. The U1 snRNP complex is responsible for pre-mRNA processing. Autoantibodies to different components of the U1 snRNP complex are associated with autoimmune conditions including systemic lupus erythematosus (SLE) and mixed connective tissue disorders (MCTD), which includes systemic sclerosis. The pattern of autoantibodies present in an individual patient may help distinguish between different connective tissue disorders. For example, patients with anti-U1A and anti-U1C IgM antibodies are more likely to have SLE than MCTD. PMID: 24158973 In addition, there is some evidence that specific antibody patterns are associated with specific symptoms. For example, among patients with SLE, the combination of anti-U1-70kD, anti-U1A and anti-U1C correlates with Raynaud's phenomenon, while anti-U1-70kD and anti-U1A (without anti-U1C) correlate with leukopenia. Another study found an association between the number of specific positive antibodies within the anti-U1 snRNP (rather than the presence of particular markers) and certain clinical features. For example, patients with three or more positive autoantibodies were more likely to have features of systemic sclerosis and less likely to have renal disease compared to patients with fewer types of antibodies.
