Nuclear antibody patterns are distinguishable fluorescent designs that occur when autoantibodies in patient serum bind with mammalian cells that have been fixed to a glass slide. The bound autoantibodies are then coated with anti-human IgG antibodies that are tagged with a fluorochrome stain. The slide is then viewed under a fluorescent microscope where the various patterns of fluorescence can be seen depending upon what autoantibodies are present. The fluorescent patterns typically reported include nuclear, centromere, or nucleolar. The nuclear pattern is more specifically described as speckled, homogenous, or peripheral. The patterns seen are associated with the autoantibodies that are produced in certain systemic autoimmune diseases including systemic lupus erythematosus (SLE), scleroderma (SSc), polymyositis/dermatomyositis (PM/DM), and rheumatoid arthritis (RA). The autoantibodies associated with these autoimmune diseases bind with antigens that are present in all cells of the body and destroy tissue in various organs. Many of the antibodies are present in more than one disorder and in differing titers. PMID: 19277826
