PRRSV strain identified (e.g. IngelvacMLV like Wild Type) in isolate by sequencing. The submitter's lab performs reverse transcriptase PCR on various specimens, including tissue, serum, oral fluid, and semen, to detect and isolate the ORF5 gene. PCR products are then used for sequencing and identifying the PRRSV strain.

Sequencing is a method used to determine the sequence of individual genes, larger genetic regions (i.e. clusters of genes or operons), full chromosomes or entire genomes. Historically, most sequencing has been performed using the chain termination method developed by Frederick Sanger in 1977. PMID: 271968 Sequencing technologies have improved dramatically, making them cheaper, faster, and more accurate. Next-generation sequencing (NGS), also known as high-throughput sequencing, deep sequencing, and second-generation sequencing, is a type of technology that uses parallel sequencing of multiple small fragments of DNA to determine sequence. This "high-throughput" technology has increased the speed and amount of DNA sequenced at a significantly reduced cost. Third-generation sequencing is another methodology currently under development that uses parallel sequencing similar to NGS. Porcine reproductive and respiratory syndrome virus (PRRSV), also referred to as Blue-Ear Pig disease, causes significant disease and can lead to reproductive failures such as weak fetuses, mummified fetuses, abortions or still borns. PRRSV is an RNA virus and belongs to the order Nidovirales, family Arteriviridae and the genus Arterivirus. PRRSV is spread among all ages of pigs and herds through direct and indirect contact. PRRSV is considered one of the most economically important infectious agents in the swine industry worldwide. The continual changing genomic characteristics of the various strains of this virus make testing of herds an important part of disease management. PMID: 26646630
