Cefazolin, also known as cefazoline or cephazolin, is a antibiotic used for the treatment of a number of bacterial infections and usually administered by either intramuscular injection or intravenous infusion. As a first-generation cephalosporin antibiotic, cefazolin is very active against gram-positive bacteria and some gram-negative bacteria. The broad spectrum of activity can be contributed to the improved stability to many bacterial beta-lactamases compared to penicillins. Due to possible cross-reactivity with penicillins, people who have experienced anaphylaxis to penicillins are not advised to receive cephalosporins. Common side effects include nausea, vomiting, diarrhea, and rash. Used in a variety of infections provided that susceptible organisms are involved, cefazolin is indicated for use in the following infections: respiratory tract, urinary tract, skin, biliary tract, bone and joint, genital, sepsis, and endocarditis. Cefazolin is on the World Health Organization's List of Essential Medicines.[Wikipedia: Cefazolin]

LP207910-3 free DRUG/TOX
The measurement of free (unbound) drug concentrations in plasma is important for certain drugs and has two advantages over measurement of total concentration: (1) the free concentration is independent of changes in plasma binding (e.g., due to changes in albumin), and (2) the free concentration is more directly related to both the efficacy and toxicity of the drug PMID: 6636249. For most drugs and the majority of patients, measuring the free and total concentrations is not necessary. However, for certain drugs, including some anticonvulsants and antibiotics, and specific patient populations, such as critically ill or immunosuppressed patients, the free concentration is a better indicator of therapeutic effect and potential toxicity.
