The Porcine Reproductive and Respiratory Syndrome Virus (PRRSv) genome contains at least eight opens reading frames (ORFs). ORF 5 contains about 600bp and is the least conserved region of PRRSV. Samples taken from infected pigs, as well as virus isolates, can be tested to determine the genetic sequence of the PRRSv. Examining the portion of the RNA that is most likely to mutate helps in determining how closely linked one PRRSv is to another, and can also be used to differentiate field strains from vaccine strains. This assay targets the ORF5 gene for Ingelvac MLV like PRRSV strain. Sample types for this test may include serum, fresh tissue, oral fluids or swabs, nasal swabs and environmental specimens like feed or feed pre-mix.

The ORF5 gene of the porcine reproductive and respiratory virus (PRRS) has been found to be extremely variable, showing polymorphism in both the nucleic acid and amino acid positions of the gene. This variability has made it a specific target for vaccine development as well as epidemiological investigation. PMID: 10640555
Studies have shown that the genetic diversity is at least as high in the European genotype as in the North American genotype and the diversity of European type PRRSV has a marked geographical pattern, with exceptionally high genetic diversity in Italian sequences.
