Term Description
The IFA method for antibody testing of Porcine reproductive and respiratory syndrome (PRRS) is fast and less expensive than other methods but can result in false positives so should be confirmed by virus isolation (VI) or RT-PCR methods. This economically important pandemic disease causes reproductive failure in breeding stock and respiratory tract illness and has shown differences in virulence, tissue trophism and replication capabilities among strains.
PRRSV has two genotypes - the North American strain (type 2 PRRSv, with the isolate prototype VR2332) and the European strain (type 1 PRRSv, with the isolate prototype "Lelystad"). Both types were discovered in their local regions around 1990 but they now share worldwide distributions. These two main genotypes differ genetically by approximately 40%. Within each genotype there is nucleotide divergence of up to 30% (Leung et al., 2010). The majority of the nucleotides that make up the two are the same but diversity within each genotype is high with a significant geographic pattern for the various types. The use of sequence information to compare PRRSV isolates is a valuable tool to monitor PRRSV strains both globally and within a farm over time. It can differentiate between resident and newly introduced viruses as well as between vaccine and field strains.

Antibody can be detected in experimenally infected pigs 7 to 10 days post inoculation. Serum samples are screened at a 1:20 dilution. (USDA).
