Cells.chromosome 3 monosomy/Cells counted in Cancer specimen by FISH
Active
Term Description
The percent of cells in the cancer specimen containing a single copy of chromosome 3 (monosomy 3) can be detected by FISH based on the number of chromosome 3 probe signals. This term is used to report the number of cells that have a chromosome 3 deletion out of total number of cells examined (e.g. 80 or of 100 cells, or 80%). Results are also typically reported in ISCN (International System for Human Cytogenetic Nomenclature) format [LOINC: 62356-1]. This term was created for, but not limited in use to, the submitter's assay which uses the Vysis chromosome 3 CEP (D3Z1) probe and a laboratory-developed BCL6 (3q27.2) probe.

Ocular melanoma is a rare cancer of the pigment cells in the eye. The most common ocular melanoma of adults is uveal melanoma. The uvea is the middle layer of the eye between the sclera and the retina. Uveal melanoma can be difficult to treat due to the lack of early signs or symptoms of disease. (https://www.aao.org/eye-health/diseases/ocular-melanoma-diagnosis) The incidence of uveal melanoma in the adult Caucasian population is about 0.7 cases per 100,000 per year. Due to the lack of early signs and symptoms and consequent delay in treatment, metastatic disease is common and occurs in up to half of all patients. In one study of patients with uveal melanoma, 62% of the melanoma specimens were positive for chromosome 3 monosomy (monosomy 3), and the finding of monosomy 3 was correlated with both the likelihood of metastatic disease as well as decreased survival. The underlying reason for the association between chromosome 3 monosomy and metastases is not yet understood. PMID: 18234992

LP62864-1 FISH
FISH (fluorescence in situ hybridization) is a cytogenetic technique used to detect and localize the presence or absence of specific DNA sequences on chromosomes. FISH uses fluorescent probes that bind to only those parts of the chromosome with which they show a high degree of sequence similarity. Fluorescence microscopy can be used to find out where the fluorescent probe bound to the chromosomes. FISH is often used for finding specific features in DNA for use in genetic counseling, medicine, and species identification. FISH can also be used to detect and localize specific mRNAs within tissue samples. In this context, it can help define the spatial-temporal patterns of gene expression within cells and tissues.
