The auramine O fluorochrome stain is a histological technique used to detect acid-fast bacilli (AFB) (e.g. Mycobacterium) using fluorescence microscopy. The test is may be performed to detect tuberculosis (TB). In auramine O stains, organisms stain bright golden yellow and non-specific debris stain pale yellow.[WHO: tb/laboratory/mycobacteriology-laboratory-manual.pdf] Fluorochrome stains are more sensitive than the Ziehl-Neelsen (ZN) method because the organisms stain brighter and the smear can be examined under a lower magnification, allowing more fields to be read in the same amount of time.
