Part Description
LP249724-8 Porcine circovirus type 2 RFLP pattern
Porcine circovirus 2 (PCV2) is a small, nonenveloped DNA virus that is associated with disease in pigs. PCV2 isolate typing is important for epidemiologic purposes, both within a single farm, regionally, and globally. Restriction fragment length polymorphism (RFLP) analysis is used to determine the "cutting pattern" for an isolate based on three different enzymes with known patterns. As a hypothetical example, one enzyme may be known to cut a specific 500 base pair (bp) segment of DNA into two pieces that are 200 and 300 bp, or three pieces that are 50, 150 and 300 bp, or to not cut the segment at all depending on the nucleotide sequence in that specific sample. The two piece pattern might be labeled "1", the three piece pattern "2", and the single, original piece "3". The individual pattern numbers for three different enzymes are put together as a single RFLP pattern, e.g., 3-1-2 or 1-1-1. Each of these RFLP patterns identifies a unique strain of PCV2, which information is used for epidemiologic analysis such as determining the source of an outbreak and tracking the spread of disease.
