This panel is used to evaluate patients who have clinical features associated with IPEX (immune dysregulation, polyendocrinopathy, enteropathy, X-linked inheritance) and primary immunodeficiencies, autoimmune disease, allergy and asthma, and graft-vs-host disease after hematopoietic stem cell transplantation.

Regulatory T-cells (Tregs) are a subset of CD4+ T-cells that provide protection from excess inflammation and tissue damage that would result from untethered immune responses directed against self, dietary, or environmental antigens. Tregs are characterized and dependent upon the expression of lineage-specific transcription factor Foxp3 and high levels of interleukin-2 (IL2) receptor alpha-chain (CD25). Tregs require the expression of CD25 for their differentiation and proliferative fitness and are dependent on IL2 signaling for their maintenance. Tregs suppress immune-mediated inflammation that is associated with autoimmune and auto-inflammatory disorders, allergies, acute and chronic infections, and cancer. Associations of Tregs with disease has prompted investigations into the manipulation of Tregs for therapeutic purposes. PMID: 27590281
