Descriptions

Aldolase is an enzyme which catalyzes the aldol reaction. It beaks down fructose 1,6-bisphosphate (F-1,6-BP) into glyceraldehyde 3-phosphate and dihydroxyacetone phosphate (DHAP), as part of glycolysis. In mammalian aldolase the key catalytic amino acid residues involved in the reaction are lysine and tyrosine. Tyrosine acts as an efficient hydrogen acceptor whereas the lysine covalently binds and stabilized the intermediates. Many bacteria use two magnesium ions in place of the lysine. Aldolase glycolytic enzyme that splits fructose 1,6-diphosphate. It is present most significantly in skeletal and heart muscle, although all cells contains samll amount, and the liver has a moderate concentration. Damage to skeletal muscle produces high levels of aldolase; this is particularly so in the case of progressive muscular dystrophy in which levels may be 10-15 times normal. There is no aldolase elevation in muscle diseases of neural origin such as neurogenic muscular atrophy, poliomyelitis, myasthenia gravis and multiple sclerosis, but any of the inflammatory conditions involving muscle may cause moderately elevated values. Used for the differential diagnosis of Duchenne's muscular dystrophy and in carriers lesser degrees of elevation in dermatomyositis, polynyositis, and limb-girdle dystrophy.
