Hepatocyte growth factor (HGF) is secreted by mesenchymal cells and acts as a cytokine, mainly on cells of epithelial origin, to regulate cell growth, motility, and morphogenesis. It plays a key role in angiogenesis, tumorogenesis, and tissue regeneration. HGF is secreted as an inactive polypeptide and is cleaved by serine proteases into a 69-kDa alpha-chain and 34-kDa beta-chain. A disulfide bond between the alpha and beta chains produces the active molecule that binds to the c-Met receptor and activates a tyrosine kinase signaling cascade. Mutations in the HGF gene are associated with autosomal recessive deafness-39, a non-syndromic hearing loss. [NCBI Gene ID: 3082] HGF plasmid DNA therapy targeted to cardiomyocytes is being examined as a potential treatment for coronary artery disease and for damaged cardiac tissue post-myocardial infarction. HGF levels may also be a marker for prognosis in Chikungunya virus-induced arthralgia, where higher HGF levels correlate with higher rates of recovery.
