Description
Hermansky-Pudlak syndrome (HPS) is associated with bleeding due to platelet dysfunction, which manifests by an absence of platelet dense bodies. Dense bodies are needed for the second phase of platelet aggregation, and in HPS, the granulophysin and CD63 components of dense bodies is lacking. The high calcium content of dense bodies allows their visualization in unfixed, unstained whole mount preparations, and platelet examination for dense bodies by whole mount electron microscopy is the gold standard for diagnosing HPS.
