Description
L-serine (L-ser) is an amino acid that plays a central role in cellular proliferation and is the major source of one-carbon groups for the de novo synthesis of purine nucleotides and deoxythymidine monophosphate. L-ser is converted to D-ser by serine racemase. Altered CNS D-ser levels have been associated with a variety of neurologic conditions, including schizophrenia and Parkinson's disease. L-ser also plays a specific role in the CNS, and patients with defects of L-ser synthesis have significant neurological impairment. Two enzyme defects linked to L-ser synthesis have been discovered: 3-phosphoglycerate dehydrogenase (3-PGDH) deficiency and phosphoserine phosphatase (PSP) deficiency. Patients with 3-PGDH deficiency can have significant neurologic impairment, including congenital microcephaly and seizures associated with decreased white matter volume and myelination, and L-ser supplementation has been used to treat the seizures in this patient population.
