MAOA (monoamine oxidase A) gene is located on the X chromosome and produces the monoamine oxidase A (MAO-A), an enzyme that degrades amine neurotransmitters, such as dopamine, norepinephrine, and serotonin. PMCID: PMC1683299 A polymorphism located upstream of the MAOA gene contains a 30 base pair variable number of tandem repeats sequence (VNTR) with 2, 3, 3.5, 4, or 5 repeated copies. The polymorphism has been shown to affect the transcriptional activity of the MAOA gene promoter by gene fusion. Transcription of the 2 or 3-repeat (short) alleles results in reduced MAOA activity and consequently increased levels of serotonin and other amine neurotransmitters, which is thought to increase the risk for aggression and psychiatric disorders. PMCID: PMC4377966 Alleles consisting of 3.5 or 4 repeats have shown higher MAOA gene expression and activity than those with 3 repeats. PMID: 17692293 Disorders associated with the upstream VNTR (uVNTR) genotype and resulting MAO-A enzyme activity include Alzheimer's disease, aggression, panic disorder, bipolar affective disorder, major depressive disorder, and attention deficit hyperactivity disorder.
