Description
Autoimmune lymphoproliferative syndrome (ALPS) is a disorder that occurs when a mutation in the FAS gene prevents the normal death cycle of the lymphocytic white blood cells. The abnormal accumulation of non-malignant lymphocytes can lead to splenomegaly, lymphadenopathy, an increased risk of malignancy, and an autoimmune cytopenia resulting in anemia and thrombocytopenia. In ALPS there is an increase in the CD4- CD8- TCR alpha beta+ T cells, FAS ligand (FAS-L), and IL-10 protein. CD4-CD8- T cells are also known as double negative T cells (DNT). The expression of an atypical isoform of CD45 called CD45R (also known as B220) on CD4-CD8-TCR alpha beta+ T cells is sensitive and specific for ALPS.
