Paraneoplastic syndromes (PNS) are diseases associated with remote effects of tumors, of which small-cell lung cancer (SCLC), ovarian cancer, breast cancer, thymoma, Hodgkin lymphoma, and testicular cancer are the most common. Stiff-person syndrome (SPS) is a rare neurological disorder characterized by stiffness of skeletal muscles with superimposed spasms, which can have an idiopathic or paraneoplastic component. First described in women with breast cancer and SPS, amphiphysin antibodies have been reported in patients with diverse paraneoplastic neurological disorders, and cancers. Amphiphysin antibodies are considered diagnosic for paraneoplastic neurologic syndromes and can direct the search for an occult tumor, recurrence of tumor, or second tumor. Source: ARUP Laboratories

Amphiphysin is a brain-enriched protein normally found only within the membrane of synaptic vesicles in neural tissue. Antibodies against amphiphysin (128kDa) are directed against the protein amphiphysin, and their presence plays an important diagnostic role in paraneoplastic syndromes (e.g. Lambert Eaton myasthenia syndrome, encephalomyelitis with rigidity, cerebellar syndrome, sensory neuronopathy, opsoclonus myoclonus ataxia, and Stiff-man syndrome).
