Fibroblast growth factor 23, or FGF23, is primarily produced in bone and helps regulate phosphate and vitamin D metabolism by binding to receptors on renal tubular cells. It is produced as a 251-amino acid precursor consisting of a signal peptide, N-terminal domain, and C-terminal domain. The bioactive form of FGF23 consists of the N- and C-terminal domains, after the signal peptide is removed, and is also called the "intact" form. Intact FGF23 is cleaved into N-terminal and C-terminal fragments, which are no longer active, though the C-terminal fragment may compete with intact FGF23 for renal tubule receptors and play a role in regulating overall FGF23 activity. The production of FGF23 is regulated by a variety of factors, including calcium and phosphate levels, 1,25 dihydroxyvitamin D, parathyroid hormone, and iron deficiency. Elevated FGF23 levels are associated with chronic kidney disease as well as cardiovascular disease. PMID: 24269946

FISH (fluorescence in situ hybridization) is a cytogenetic technique used to detect and localize the presence or absence of specific DNA sequences on chromosomes. FISH uses fluorescent probes that bind to only those parts of the chromosome with which they show a high degree of sequence similarity. Fluorescence microscopy can be used to find out where the fluorescent probe bound to the chromosomes. FISH is often used for finding specific features in DNA for use in genetic counseling, medicine, and species identification. FISH can also be used to detect and localize specific mRNAs within tissue samples. In this context, it can help define the spatial-temporal patterns of gene expression within cells and tissues.
