L-Proline is one of the twenty proteinogenic units used in the building blocks of proteins. The other nineteen units are all primary amino acids, but due to the cyclic binding of the three-carbon side chain to the nitrogen of the backbone, proline lacks a primary amine group (ˆ'NH2). The nitrogen in proline is referred to as a secondary amine. Proline is sometimes called an amino acid, although the IUPAC definition of an amine requires a carbon-nitrogen double bond.

Proline is an amino acid that is the cyclized form of glutamic acid. The hydroxylation of proline increases the stability of collagen and is critical for maintaining connective tissue. Increased with iminoglycinuria,Type I and II hyperprolinemia, and lysinuric protein intolerance.

Creatinine or creatine anhydride, is a breakdown product of creatine phosphate in muscle. The loss of water molecule from creatine results in the formation of creatinine. It is transferred to the kidneys by blood plasma, whereupon it is eliminated by glomular filtration and partial tubular excretion. Creatinine is usually produced at a fairly constant rate and measuring its serum level is a simple test. A rise in blood creatinine levels is observed only with marked damage to functioning nephrons; therefore this test is not suitable for detecting early kidney disease. Creatine and creatinine are metabolized in the kidneys, muscle, liver and pancreas.
