An enzyme that catalyzes the hydrolysis of adenosine to inosine with the elimination of ammonia. Since there are wide tissue and species variations in the enzyme, it has been used as a tool in the study of human and animal genetics and in medical diagnosis. EC 3.5.4.4.

Adenosine deaminase (ADA) is a purine salvage pathway enzyme, which catalyzes the irreversible deamination of adenosine into inosine and ammonia. Elevated adenosine deaminase activities are found in tuberculous pleural and pericardial effusions. In addition, increased ADA levels in cerebrospinal fluid (CSF) have been observed in tuberculous meningitis. These ADA increases are caused by the cellular immune response in the fluid compartment, especially in the activation of T lymphocytes. Thus, many disorders can cause elevations. Deficency of adenosine deaminase is the most frequent known cause of SCID (Severe combined immunodeficency disease) it is observed in 25% of cases.
