Keyhole limpet hemocyanin (KLH) is a large, multisubunit, oxygen-carrying, metalloprotein found in the hemolymph of the giant keyhole limpet, Megathura crenulata. KLH is being used in a variety of cancer vaccines, including non-Hodgkins lymphoma, cutaneous melanoma, breast and bladder cancer. These vaccines contain specific tumor-associated antigens conjugated to KLH to stimulate anti-tumor immune responses which can destroy tumor cells. KLH is very immunogenic, so assays to monitor humoral immune responses against KLH in human serum have been developed. An individual's humoral and cellular anti-KLH response is indicative for their immunocompetence (Grant et al. 2008, PMID: 18295445. Testing for anti-KLH antibodies may also be performed as a biomarker for vaccine-immunogenicity of KLH-containing and KLH-conjugated vaccines (Irfan et al. 2007, PMID: 17936425.
