Real-time PCR is performed to detect the presence of MERS-CoV RNA in respiratory specimens (nasopharyngeal, oropharyngeal swabs, sputum, lower respiratory aspirates/washes), serum, and stool. This code was created for, but not limited in use to, the submitter's primer and probe set, NCV-2012 rRT-PCR (CDC; Catalog #KT0136), which targets the nucleocapsid protein gene (NCV.N2 and NCV.N3) and upstream the MERS-CoV envelope protein gene (NCV.upE).

The Middle East respiratory syndrome coronavirus (MERS-CoV), also termed NCV-2012, EMC/2012 or SARS-like virus, is a Betacoronavirus, a novel coronavirus (nCoV). Coronaviruses commonly cause respiratory illnesses, including the common cold, but the MERS-CoV is more lethal. The first MERS-CoV case was reported in September 2012 in the Middle East. Testing is currently (2013) restricted to patients exhibiting the clinical symptoms of MERS-CoV infection and having the epidemiological risk factors. Several highly sensitive, confirmatory real-time RT-PCR assays exist for rapid identification of MERS-CoV from patient samples (such as bronchoalveolar lavage or sputum). Targets include upE (targets elements upstream of the E gene), 1A (targets the ORF1a gene) and nucleocapsid protein genes (specific to MERS-CoV).
