Ornithine/Citrulline is a marker for gyrate atrophy of the retina, ornithine transcarbamylase deficiency, carbamoyl-phosphate synthase deficiency, and hyperornithinemia-hyperammonemia-homocitrullinuria syndrome, which are all conditions that some states include on their newborn screening panel.

LP15753-4 Ornithine
Ornithine is an amino acid that, under the action of the enzyme arginase on L-arginine, yields urea. Therefore, ornithine is a central part of the urea cycle, which eliminates excess nitrogen. Ornithine is not an amino acid coded by DNA, and in that sense, it is not involved in protein synthesis. In mammalian non-hepatic tissues, however, the urea cycle is mainly involved in arginine biosynthesis, making ornithine an important metabolic process. It is not directly a product of the the genetic code because polypeptides containing unprotected ornithines undergo spontaneous lactamization. This proved to be a problem when ornithine was originally classified among the other 20 naturally occuring amino acids.
